Wnt Activity and Cell Proliferation Are Coupled to Extracellular Vesicle Release in Multiple Organoid Models

Extracellular vesicles (EV) are considered as a potential tool for early disease diagnosis; however, factors modifying EV release remain partially unknown. By using patient-derived organoids that capture the cellular heterogeneity of epithelial tissues, here we studied connection between Wnt-producing microniche and secretion in multiple tissues. Although nearly all cells pancreatic ductal (PD) adenocarcinoma (PDAC) samples expressed porcupine (PORCN), an enzyme critical Wnt secretion, only subpopulation lung bronchiolar (NL) (LUAD) organoid produced active Wnt. The proliferating was shaped not by PORCN + NL LUAD but also fibroblast-derived EVs. This effect could be blocked inhibitors. Whereas inhibiting PD or critically changed both cell proliferation release, these were uncoupled from each other PDAC. Sorting CD133 identified population with high percentage elevated which may explain marks malignant behavior. Collectively, show rate, induced pathway activation, is coupled to higher finding when developing EV-based diagnostic tools.
